Advanced Light Microscopy - Imaging Theory

FSI-9MIA Acad. year: 2023/2024 Summer semester

The course explains principles of light microscopy of three-dimensional objects in terms of wave optics. Image properties are described using the formalism of 3D pupil and transfer functions. Properties of advanced microscopic techniques for 3D imaging are explained, especially those of confocal microscopy and coherence-controlled holographic microscopy.

Entry knowledge

Wave and Fourier optics, geometrical optics, fundamentals of light microscopy.

Rules for evaluation and completion of the course

Examination is oral. Detailed knowledge of explained matter is proved. Examined student can use a literature.
Presence at lectures is not obligatory, but recommended.

Aims

Understanding of principles of 3D light microscopy and microscopic imaging.
Acquiring of methods for fundamental 3D imaging characteristics calculation.
Knowledge of fundamentals of theoretical description of microscope imaging of 3D objects of their relation to experiment and competence to apply them in practice.
